Data pending verification. Identity data in the specification below has been compiled for pre-launch and is awaiting verification against supplier documentation and batch COAs. Highlighted fields are unconfirmed.
Note: Recombinant protein: identity, expression system, purity method and endotoxin level must be confirmed from the supplier COA. The supplier's '≥99%' purity specification needs checking for a protein product. Purity specification to be confirmed against the supplier COA.

ACE-031 overview

ACE-031 is a recombinant fusion protein made of the extracellular domain of human activin receptor type IIB linked to the Fc region of human IgG1. It is supplied as a lyophilised solid for in-vitro laboratory research.

ACE-031 research background

Clinical trials – mixed results

Evidence status: Findings summarised here include controlled human trials with mixed or negative results, alongside preclinical research. They are not established effects in humans, have not been evaluated by the MHRA, and are provided for scientific context only. This product is supplied strictly for laboratory research and is not for human or veterinary use.

Overview & discovery history
  • ACE-031 (ramatercept) is a recombinant fusion protein, not a short peptide. It combines the extracellular domain of activin receptor type IIB (ActRIIB) with a human IgG1 Fc fragment, and acts as a ligand trap for myostatin and related TGF-β-family ligands. [1][2].

Mechanisms investigated
  • Sequestration of myostatin and activins, reducing ActRIIB signalling in muscle [1].
Research areas & model types
Research areaModel typeSummary
Skeletal muscle massMouse[1]
Muscle mass and strengthNon-human primate (marmoset)[4]
Healthy postmenopausal womenHuman (Phase 1)[2]
Duchenne muscular dystrophyHuman (Phase 2 RCT)[3]
Human clinical research status
  • A Phase 2 trial in boys with Duchenne muscular dystrophy was discontinued; non-muscle adverse events contributed to the decision [3]. ACE-031 is not an approved medicine.
Safety observations reported in studies
  • The single-dose Phase 1 study reported it as generally well tolerated [2]. In the DMD trial, non-muscle adverse events contributed to stopping the study [3].
Known limitations
  • Clinical development was discontinued.
Storage & handling for laboratory use

Store at −20 °C. Protect from light.

Store lyophilised material sealed at −20 °C or below, protected from light and moisture. Allow the vial to equilibrate to room temperature before opening to limit condensation.

  • Allow the sealed vial to reach room temperature before opening to limit condensation.
  • Any solutions should be prepared only for in-vitro laboratory work under your own validated protocol; aliquoting limits repeated freeze–thaw cycles.
  • Handle with standard laboratory PPE in line with your risk assessment and the Safety Data Sheet (SDS). Britpep does not provide preparation or administration guidance.

Emerging & theoretical

Unconfirmed — hypotheses & ongoing research

These are unconfirmed hypotheses and early-stage lines of enquiry, not established effects. Not evaluated by the MHRA. Not for human or veterinary use.

Hypotheses proposed by researchers

  • Researchers continue to propose ActRIIB-pathway inhibition as a strategy in neuromuscular research [3][4].

Preprints: none included (no relevant preprint could be verified). Anecdotal, user-reported or social-media material is deliberately excluded.
